研究方向
本課題組的研究興趣為建立方法與手段改造蛋白質的穩定性,解析與
蛋白質摺疊密切相關的一類蛋白——
分子伴侶蛋白的結構與作用機制,並利用所建立的
蛋白質穩定性最佳化平台研究一些重要的基礎科學問題,如參與表觀遺傳調控的一類組蛋白甲基轉移酶的活化機制。 我們將綜合利用細菌遺傳學、分子生物學、生物化學、生物物理、生物信息學與結構生物學等手段,開展如下幾方面的研究工作:
1.針對複雜蛋白質樣品難以獲得、不穩定的瓶頸問題,發展新型蛋白質體內穩定性檢測探針,打造提升
蛋白質穩定性的高通量篩選平台,研發新型高效異源蛋白質過表達體系,最佳化一些工業上、醫藥上迫切需要的蛋白質的穩定性。
2.發現與鑑定新型
分子伴侶蛋白,闡釋其作用機理,並藉助結構生物學手段研究分子伴侶蛋白和底物的相互作用、解析並表征體內蛋白質穩態網路。
3.篩選針對
澱粉樣蛋白的多肽抑制劑,為預防及治療神經退行性疾病、
二型糖尿病等蛋白質錯誤摺疊引起的疾病提供新的思路。
4.以提升組蛋白H3K4甲基轉移酶家族的穩定性為切入點,解析結構靈活性與其催化活性間的關係,闡述其被其他蛋白質複合物激活的機制,為以此家族蛋白為靶點的藥物設計提供新思路。
主要成就
長期聚焦蛋白質摺疊這一重要科學問題,圍繞分子伴侶及酶作用機制、蛋白質穩定性最佳化等展開基礎及套用性基礎研究。發展了篩選與鑑定分子伴侶的新策略,闡明了ATP非依賴型分子伴侶獨特的作用機制;建立了蛋白質穩定性進化的系統性方法,在提升酶穩定性與減少澱粉樣蛋白聚集方面展現了良好的套用前景。代表性成果以一作或通訊作者身份發表在Nature Structural & Molecular Biology,PNAS,Nature Communications,eLife等期刊上。
