2004年12月28日在上海逝世，享年80歲。何惠鑒本來住在美國匹茲堡。 因擔任上海博物館客座館長，所以暫居上海。何的家人說，死因是糖尿病引發感染。 何惠鑒1947年畢業於嶺南大學。他受業於著名史學家陳寅恪，畢業論文也是由陳指導。 1950年，陳寅恪秘密安排把何惠鑒送去哈佛攻讀藝術史。 1953年，何在哈佛獲得中國史與亞洲藝術雙碩士學位。 1959至1983年擔任克利夫蘭「東方與中國藝術美術館」館長。 1984至1994年擔任Nel-son-Atkins博物館的顧問。 50年代期間，西方學者對中國繪畫研究甚少，總覺得十分費解，創作年代更難以查考。 但何惠鑒利用他「藝術偵探」的本事和深度的研究，引領了西方學者的研究之風。 何惠鑒在中國繪畫和佛教美術方面堪稱權威。 著作有《元代文人畫序說》、《董其昌的世紀》（合著）等。 詳盡完備的The Century of Tung Ch'i-ch'ang (1555–1636)獲得了首屆Shimada獎。
